چکیده

Variants in Atherosclerosis, Obesity, and Bone Traits Since learning of the Letter to the Editor by Lyons1 we sequenced the 5-lipoxygenase (5-LO) gene in strains 129/SvJ, CAST/EiJ, and a congenic strain carrying a CAST/Ei allele. The sequences are in agreement with those indicated by Dr Lyons. CAST is a “wild-derived” strain, and we can only speculate that the stock we sequenced may not have been fully inbred; we obtained our stocks in or around 1990 from the Jackson Laboratory. When we reported the 5-LO amino acid differences between CAST and B6, we were unaware of their effect on enzymatic activity that was subsequently reported by Kuhn et al.2 Because we based our conclusions in our article solely on the low transcript and protein levels observed in congenic mice carrying the CAST 5-LO allele, the validity of our results are not altered. Overall, our studies in mice and humans as well as more recent ones from other groups are consistent with the proatherogenic role that 5-LO and the leukotriene pathway play in cardiovascular disease.

5‐Lipoxygenase Gene Variants Are Not Associated With Atherosclerosis or Incident Coronary Heart Disease in the Multi‐Ethnic Study of Atherosclerosis Cohort

BACKGROUND The arachidonate 5-lipoxygenase enzyme plays a crucial role in mediating inflammation to maintain homeostasis, yet certain allelic variants of the 5-lipoxygenase gene, ALOX5, may increase risk of atherosclerosis and coronary heart disease (CHD). Further, relations between ALOX5 and disease outcomes may be enhanced or attenuated depending on the bioavailability of 5-lipoxygenase enzym...

Arachidonate 5-lipoxygenase variants in atherosclerosis, obesity, and bone traits.

Atherosclerosis, Obesity, and Bone Traits To the Editor: I write regarding publications by Mehrabian et al1,2 appearing in Circulation Research and a subsequent publication by Kuhn et al3 appearing in Arteriosclerosis, Thrombosis, and Vascular Biology. The key implication from the combination of these reports was that amino acid substitutions in arachidonate 5-lipoxygenase (ALOX5) could account...

Variants of the arachidonate 5-lipoxygenase-activating protein (ALOX5AP) gene and risk of ischemic stroke in Han Chinese of eastern China☆

Variants of the arachidonate 5-lipoxygenase-activating protein (ALOX5AP) gene have been suggested to play an important role in the pathogenesis of atherosclerosis and ischemic stroke. This study was aimed to explore the association of ALOX5AP variants with ischemic stroke risk in Han Chinese of eastern China. A total of 690 ischemic stroke cases and 767 controls were recruited. The subjects wer...

Human Lipoxygenase Pathway Gene Variation and Association with Markers of Subclinical Atherosclerosis in the Diabetes Heart Study

AIMS Genes of the 5-lipoxygenase pathway are compelling candidates for atherosclerosis. We hypothesize that polymorphisms in ALOX12, ALOX15, ALOX5, and ALOX5AP genes are associated with subclinical atherosclerosis in multiple vascular beds. METHODS Families with two or more siblings with type 2 diabetes and their nondiabetic siblings were studied as part of the Diabetes Heart Study (DHS). Eur...

5-Lipoxygenase-activating protein: a potential link between innate and adaptive immunity in atherosclerosis and adipose tissue inflammation.

Transforming growth factor-beta (TGF-beta) is a major antiinflammatory mediator in atherosclerosis. Transgenic ApoE(-/-) mice with a dominant-negative TGFbeta type II receptor (dnTGFbetaRII) on CD4(+) and CD8(+) T cells display aggravated atherosclerosis. The aim of the present study was to elucidate the mechanisms involved in this enhanced inflammatory response. Gene array analyses identified ...
